What version of Knoppix are you using?

The simple answer is, no, because a CD is read-only media. However, you can simulate a read/write media by using a persistent disk image (versions >=3.8.1). For that you'll need a CD and some read/write media, e.g. a USB key, a harddrive, etc. The complex answer is, yes, if you remaster the CD with your new settings.

I recommend using the CD + USB key approach. It's simple, versatile, and will do what you want.
