I'm a bit confused on what you want to do (restart the network or configure it?) but to restart the network you must be root and do:
/etc/init.d/networking restartconfiguring the network is another matter.

Will that save the settings and changes i made to the network .conf files?

It shouldn't affect the conf files at all, just restart the daemons / scripts.

You're right, the Knoppix disc isn't writeable. That means you'll have to use some other medium like a floppy disk or the hard drive to save your data to. In most cases you can mount a floppy disk by doing (IIRC):
mount /floppyand then saving your data to the /floppy/ directory.
