Skypes linux forum section has a couple of good stickies on this which you might want to look at: http://forum.skype.com/viewforum.php?f=18
I recently installed Knoppix 3.8.1 on my hard drive and it's generally wonderful. I had Skype working on another Linux box with Mandrake 10.0.
I installed Skype from their Debian package and it works fine apart from not having any sound. Other applications have working sound and the microphone is getting through to the speakers, but I have not yet tried recording sound from it.
The system is a Duron 1200 on an Asus A7N266-VM with the on-board graphics and sound.
Any ideas?

If you can't be bothered trawling through those skype forums then generally skype will run in 2 ways, either as:
skype
or, if the above does not give you any sound then:
artsdsp -m skype
i.e. if it doesn't work stand alone then run it under arts.
