One thing I really need for Knoppix to be closer-to-perfect is IPv6 support. It would be really cool if applications like ping6 and traceroute6 were installed by default, and that all the internet apps were IPv6 enabled.

So far, only the IPv6 kernel module is present, but that doesn't leave you with many possibilities of playing around :P
